One hour time-lapse of invasive structures (“invadosomes”, PIP2 labeling, cyan) and basement membrane (laminin::GFP, magenta) shows the anchor cell breaching the basement membrane in the left panel. Anchor cell invasion creates a breach in the basement membrane at the mid L3 larval stage and initiates contact with the underlying vulval cells. Shortly after the anchor cell breaches the basement membrane, the anchor cell expands the breach. After the gap in the basement membrane widens to the edge of the anchor cell, the gap continues to expand over the vulval cells (by sliding) until it reaches the vulD cells, where it is stabilized by integrin adhesion.
