It has been three weeks in the lab already! In this post, I am just giving a general update on the research that I described in my first post.
Two weeks ago, I included pictures of a transfection with an efficiency rate of roughly 5%; this transfection was performed using Lipofectamine LTX. Now, after using Lipofectamine 2000, we have been able to perform a transfection with an efficiency rate greater than 20%. Simultaneously, we have decided to transfect 293T cells, an easily transfectable cell line (up to 95% of cells should transfect), to test that our CRISPRs work. We will soon be screening for alleles targeted by CRIPSRs using genotyping PCR with primers that we confirmed last week. Additionally, we are looking at mitochondrial dynamics using knockout cell lines as the protein that we’re studying are believed to play a role in mitochondrial dynamics.
As the research trips along, I find myself looking forward to giving a chalk talk. I cannot wait to organize all of this information into a presentation.
