Why’s it gotta be this way? I know things have been rough between us lately, but all I’m asking is for you not to religate. Just take the insert! I’m sorry I didn’t phosphorylate it last time, but give me a second chance! I promise this time I’ll treat you better and appreciate you for the beautiful vector you are. Things will be different! I’ll let your restriction digest run for three hours and I’ll CIP you. I’m only here for two more weeks, and with each failed transformation taking two days, there isn’t much time to fix this relationship. Can’t we just forget the past and successfully transform into E.coli? There’s lots of cool stuff we could do together if you would just accept the insert and not religate. Remember all the amazing ideas we had planned for our future back when we were just a couple of kids doing theoretical ultrasensitivity modeling? Let’s make that a reality.
